Rudhof was introduced to boxing at the age of ten by his great-uncle and first trainer, the Kostheim freight forwarder Peter Krimmel. Rudhof began training as a locksmith at Opel in Rüsselsheim and trained at the local boxing club.

In 1953, he became the Hesse featherweight champion for the first time, which he won several times. After 162 amateur fights, he turned professional in 1957. He won the German lightweight title in 1959 against the Berlin title holder Rudi Langer. He defended the title in this weight class five times until 1962. During his time as a professional boxer, he had 84 fights between 1957 and 1968 (67 wins, 29 of them by knockout). Rudhof won three German championship titles in the light-welterweight, semi-welterweight and welterweight divisions. He won his first title as European champion in 1963 at lightweight against the Italian Giordano Campari in Rüsselsheim. He won his second European Championship title in 1967 in Frankfurt am Main against the Finn Olli Mäki in the super-lightweight division. Rudhof received several awards in Rüsselsheim for his great sporting achievements.

After the end of his professional career in 1968, Rudhof opened a sporting goods store in Kostheim. His friends and companions included the boxers Karl Mildenberger, Gerhard Zech, Peter Müller and Karl Furcht as well as his trainer, the former heavyweight boxer Arno Kölblin. He died shortly before a planned exhibition fight in Wiesbaden.
